The effects of seed and pericarp isolation of Radix Glehniae(aks.CoasiaI GIehnia Root)on seed germination and seedlings of Chinese cabbage were studied to explore the activity of endogenous inhibitors in seeds of Radix Glehniae.The extracts from different parts of the seeds of Radix Glehniae were separated by systematic solvent method,and the petroleum ether phase,dichloromethane phase,ethyl acetate phase,n-butanol phase,methanol phase and water phase were separated,and the germination experiments were carried out relative to Chinese cabbage seeds.The biological activity of endogenous inhibitors in Radix Glehniae seeds was determined.The results showed that:①The separation of seeds and pericarps of Radix Glehniae had a certain inhibitory effect on the germination of Chinese cabbage seeds,in which methanol phase was the strongest,followed by ethyl acetate phase.Compared with the control group,the germination index of each treatment group decreased to a certain extent,and the average germination days of methanol phase was significantly longer than that of other treatments.②The separated phases of seeds and pericarps of Radix Glehniae showed obvious differences in inhibiting root length and bud length of Chinese cabbage seedlings,among which methanol phase was the most significant,followed by ethyl acetate phase.The results were consistent with the germination index.The isolation of different parts of the seeds of Radix Glehniae had different inhibitory effects on the seed germination and seedlings of Chinese cabbage,which indicated that the seeds and pericarps of Radix Pseudostellariae contained highly active inhibitors,which mainly existed in methanol phase and ethyl acetate phase.The endogenous inhibitor in the seeds of Radix Glehniae is the main reason for its dormancy.
