Effect of Tinospora crispa extract on diabetic nephropathy mice and their acute toxicology
AIM To observe the effect of Tinospora crispa ethanol extract (TCEE) and their solvent extracts on ICR mice with diabetic nephropathy (DN) and their acute toxicities.METHOD LD50 of TCEE was determined by modified Kaber method.One hundred mice with DN induced by injection of STZ and fed high fat diet were divided equally into vehicle-treated group,the low,middle and high dose TCEE groups,EtOAc fraction group,BuOH fraction group,water soluble fraction group,Captopril group and Xiaoke Pills group,and setting the control group (n =10).At the end of 6thweek,the effects of T.crispa on serum biochemical indexes (BUN,Crea,TC,and Cys-C) and the PAS staining histomorphology of kidney tissues were evaluated.RESULTS LD50 of TCEE was 4 887 mg/kg.The levels of biochemical indexes of mice with diabetic nephropathy were significantly reduced with an improved renal function in TCEE and BuOH fraction group.PAS staining showed that the symptoms,including glomerular hypertrophy,thickening of glomerular basement membrance,and mesangial expansion,were significantly improved,especially in high dose TCEE group and BuOH fraction group.CONCLUSION T.crispa is a kind of low toxicity herb and it can reduce the level of blood glucose,total cholesterol and improve the renal function in DN mice in the safety dosage.
