毛蒟挥发油测定及其贮存期考察

目的 水蒸汽蒸馏法提取广西产毛蒟中挥发油,GC-MS法测定挥发油中β-石竹烯含有量,并对其最佳贮存期进行考察.方法 挥发油的分析色谱柱为HP-5MS (30 m×0.25 mm,0.25 μm);程序升温;载气为氦气;水杨酸甲酯为内标物.结果 挥发油得油率在0.23%~1.64%之间,贮存1年后其下降率为22.68%,1.5年后下降率为41.63%;β-石竹烯在0.033 9~0.542 4μg范围内与峰面积呈良好的线性关系(r=0.999 6),平均回收率为98.5% (RSD=2.0%,n =6);β-石竹烯含有量在31.363 8 ~338.957 2 mg/mL之间,一年后其下降率为35.03%,一年半后下降率为44.53%.结论 建议毛蒟的贮存时间不超过1年.
Determination of volatile oil in Piper puberulum and its proper storage time
AIM To determine β-caryophyllene content in volatile oil isolated from Piper puberulum by GCMS choose its proper storage time.METHODS The analysis of volatile oil was performed on HP-5MS (30 m ×0.25 mm,0.25 μm) column with temperature-programming mode,carrier gas was He,and internal standard substance was methyl salicylate.RESULTS Volatile oil rate was between 0.23% and 1.64%.A year later,its drop rate was 22.68%.After a year and a half,it was 41.63%.There was a good linear relationship for β-caryophyllene during 0.033 9-0.542 4 μg (r =0.999 6),the average recovery rate was 98.5% (RSD =2.0%,n =6),the content of β-caryophyllene was 31.363 8-338.957 2 mg/mL.A year later,its drop rate was 35.03%.After a year and a half,it was 44.53%.CONCLUSION It suggests that the storage time of P.puberulum should be no less than one year.
