摘要

目的 建立UPLC-MS/MS法同时测定百合知母汤中菝契皂苷元、王百合苷C、知母皂苷AⅢ、芒果苷、新芒果苷、王百合苷A、王百合苷B、知母皂苷BⅢ、异芒果苷、知母皂苷BⅡ的含量.方法 分析采用Agilent Poroshell 120 EC-C18色谱柱(3.0 mm×150 mm,2.7 μm);流动相水(含 0.1%甲酸)-乙腈(含 0.1%甲酸),梯度洗脱;体积流量0.5 mL/min;柱温 30℃;电喷雾离子源;正负离子扫描;多反应监测模式.结果 10 种成分在各自范围内线性关系良好(r>0.998 1),平均加样回收率 95.65%~106.5%,RSD 1.2%~5.5%.结论 该方法准确、灵敏、快速,可用于百合知母汤的质量控制.

Abstract

AIM To establish a UPLC-MS/MS method for the simultaneous content determination of sarsasapogenin,regaloside C,timosaponin A Ⅲ,mangiferin,neomangiferin,regaloside A,regaloside B,timosaponin BⅢ,isomangiferin and timosaponin BⅡ in Baihe Zhimu Decoction.METHODS The analysis was performed on a 30℃thermostatic Agilent Poroshell 120 EC-C18 column(3.0 mm×150 mm,2.7 μm),with the mobile phase comprising of water(containing 0.1%formic acid)-acetonitrile(containing 0.1%formic acid)flowing at 0.5 mL/min in a gradient elution manner,and electron spray ionization source was adopted in positive and negative ion scanning with multiple reaction monitoring mode.RESULTS Ten constituents showed good linear relationships within their own ranges(r>0.998 1),whose average recoveries were 95.65%-106.5%with the RSDs of 1.2%-5.5%.CONCLUSION This accurate,sensitive and rapid method can be used for the quality control of Baihe Zhimu Decoction.
