Effects of Yunkang Oral Liquid on postpartum recovery and offspring quality of pregnant mice with iron deficiency anemia
AIM To explore the effects of Yunkang Oral Liquid on postpartum recovery and offspring quality of pregnant mice with iron deficiency anemia(IDA).METHODS A pregnant mouse model of IDA was established by feeding of iron deficiency food combined with bloodletting.After successful modeling,ferrous succinate tablets(52 mg/kg)or Yunkang Oral Liquid(9,6,4 mL/kg)was given by gavage on the 7th day of pregnancy,once daily until 6 weeks after delivery followed by the examination of the related signs of mice(spontaneous behaviors,grasping force,back temperature,claw color);and the measurment of levels of organ indices(heart,spleen,thymus coefficient),peripheral blood indices(RBC,HGB,HCT,MCH,MCHC),and serum factors related to iron metabolism(EPO,FEP,SF,sTfR,TFR1).The mice also had their hepatic expressions of FPN and Tf proteins detected by immunohistochemistry;their hepatic protein expressions of TFR1 and Hepcidin detected by immunofluorescence;their hepatic mRNA expressions of Hepcidin,FPN,Tf and TFR1 determined by RT-qPCR;and their offspring related conditions(survival rate,spontaneous behaviors,grip strength,claw color,heart coefficient,spleen coefficient,body length,peripheral blood indices levels)measured as well.RESULTS Yunkang Oral Liquid increased the number of spontaneous activities,grasping force,back temperature and claw color of pregnant rats with IDA after delivery(P<0.05,P<0.01);reduced their heart and spleen coefficient(P<0.01);increased their levels of RBC,HGB,HCT,MCH and MCHC in whole blood(P<0.05,P<0.01);reduced their serum levels of EPO,FEP,TFR1 and sTfR(P<0.05,P<0.01),and increased their SF level(P<0.05,P<0.01);increased their hepatic mRNA expressions of Hepcidin and FPN(P<0.05,P<0.01),and reduced their mRNA and protein expressions of Tf and TFR1(P<0.05,P<0.01).In addition,compared with the offsprings of the model group,those of each Yunkang Oral Liquid group shared improved survival rate;increased number of spontaneous behaviors and grip strength(P<0.05,P<0.01);decreased levels of heart coefficient and spleen coefficient(P<0.05,P<0.01),increased body length(P<0.05,P<0.01);and increased levels of RBC,HGB and HCT in whole blood(P<0.01)as well.CONCLUSION Yunkang Oral Liquid can improve the anemia status of pregnant rats with IDA,promote their postpartum recovery and improve the quality of their offsprings.
