目的 研究乌头根际土壤真菌钩状木霉菌Trichoderma hamatum JKT-28 次生代谢产物的化学成分.方法 基于化学表观遗传修饰策略,用真菌 2 号培养基进行液体发酵获得钩状木霉菌的次生代谢产物,经乙酸乙酯萃取后,采用硅胶柱、动态轴向压缩柱、半制备液相、重结晶等方法对其进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构.结果 从中分离得到12 个化合物,分别鉴定为4-acetyl-3-hydroxy-6-methy-pyran-2-one(1)、3-methyl-4-phenyl-2(5H)-furanone(2)、6-pentyl-α-pyrone(3)、volemolide(4)、lignoren(5)、enniatin A1(6)、wickerols A(7)、demethylincisterol A3(8)、(22E)-5a,8a-epidioxyergosta-6,22-dien-3b-ol(9)、daidzein(10)、cyclo(L)-Pro-(L)-Phe(11)、Cyclo(D)-Pro-(D)-Ile(12).结论 化合物 1~12 均为首次从钩状木霉菌中得到,化合物 1 为首次从木霉属真菌中发现.
Chemical constituents from the secondary metabolites of Trichoderma hamatum in rhizosphere soil of Aconitum carmichaelii
AIM To study the chemical constituents from secondary metabolites of Trichoderma hamatum in rhizosphere soil of Aconitum carmichaelii Debx.METHODS Based on the strategy of chemical epigenetic modification,the secondary metabolites of T.hamatum were obtained by culturing in fungus No.2 medium.Compounds were separated and purified from T.hamatum ethyl acetate extract by reversed-phase column chromatography,dynamic axial compression chromatography,C18 column chromatography,semi-preparative HPLC.The structures of obtained compounds were identified by physicochemical properties and spectral data.RESULTS Twelve compounds were isolated and identified as 4-acetyl-3-hydroxy-6-methy-pyran-2-one(1),3-methyl-4-phenyl-2(5H)-furanone(2),6-pentyl-α-pyrone(3),volemolide(4),lignoren(5),enniatin A1(6),wickerols A(7),demethylincisterol A3(8),(22E)-5a,8a-epidioxyergosta-6,22-dien-3b-ol(9),daidzein(10),cyclo(L)-Pro-(L)-Phe(11),Cyclo(D)-Pro-(D)-Ile(12).CONCLUSION Compounds 1-12 are obtained from T.hamatum for the first time,and compound 1 is found from Trichoderma genus for the first time.
