簕欓花椒化学成分及其抑菌活性研究

目的 研究簕欓花椒Zanthoxylum avicennae(Lam.)DC.的化学成分及其抑菌活性.方法 簕欓花椒95% 乙醇提取物采用硅胶、Sephadex LH-20、ODS、TLC及半制备HPLC进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构.采用微量稀释法测定各化合物对耐甲氧西林金黄色葡萄球菌(MRSA)生长的抑制作用.结果 从中分离得到15个化合物,分别鉴定为木栓酮(1)、linoleic glyceride(2)、b-friedoolean-5-ene-3-β-ol(3)、(±)-蛇菰脂醛素(4)、(10E,12Z)-9-羟基-10,12-亚油酸乙酯(5)、(9Z,12Z,14E)-16-羟基-9,12,14-亚油酸乙酯(6)、(S)-24-麦角甾-4-胆甾烯-3-酮(7)、β-谷甾醇(8)、avicennol ethyl ether(9)、羟基酪醇(10)、6,7-二甲氧基香豆素(11)、(R)-(-)-mellein methyl ether(12)、(-)-二氢欧山芹素(13)、avicennol methyl ether(14)、(Z)-avicennone(15).化合物3、5、9、14~15的MIC值为250μg/mL,化合物1~2、4、6~8的MIC值为125μg/mL,化合物13的MIC值为31.25μg/mL.结论 化合物1~6为首次从花椒属植物中分离得到,化合物7~13为首次从该植物中分离得到,另外首次报道了化合物9、14的碳谱数据.化合物1~9、13~15均有抑菌活性,其中化合物13的抑菌活性较强.
Chemical constituents from Zanthoxylum avicennae and their antibacterial activities
AIM To study the chemical consitituents from Zanthoxylum avicennae (Lam.) DC.and their antibacterial activities.METHODS The 95% ethanol extract from Z.avicennae was isolated and purified by silica gel,Sephadex LH-20,ODS,TLC and semi-preparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.Microdilution broth method was employed to the antibacterial activities of the compounds on methicillin-resistant Staphylococcus aureus (MRSA).RESULTS Fifteen compounds were isolated and identified as friedelin (1),linoleic glyceride (2),b-friedoolean-5-ene-3-β-ol (3),(±)-balanophonin (4),(10E,12Z)-9-hydroxy-10,12-octadecadienoic acid ethyl ester (5),(9Z,12Z,14E)-16-hydroxy-9,12,14-octadecatrienoic acid ethyl ester (6),(S)-24-ergost-4-en-3-one (7),24-propylcholesterol (8),avicennol ethyl ether (9),hydroxytyrosol (10),6,7-dimethylcoumarin (11),(R)-(-)-mellein methyl ether (12),(-)-columbianetin (13),avicennol methyl ether (14),(Z)-avicennone (15).The MIC values of compounds 3,5,9,14-15 were 250μg/mL,the MIC values of compounds 1-2,4,6-8 were 125 μg/mL,and the MIC value of compound 13 was 31.25 μg/mL.CONCLUSION Compounds 1-6 are isolated from genus Zanthoxylum for the first time.Compounds 7-13 are isolated from this plant for the first time.In addition,the carbon spectrum data of compounds 9and 14 are reported for the first time.Compounds 1-9 and 13-15 have antibacterial activities,and compound 13 has strong antibacterial activity.
