为鉴定不同欧李种质资源的S基因型,分析欧李S基因序列特点,以70份欧李种质为材料,通过PCR特异性扩增S基因,并克隆测序得到其基因序列.结果表明:利用BFP93/BFP94-1引物,在52份种质中各鉴定到2个S基因,在16份种质中各鉴定到单个S基因,在3-32-扁黄和10-32两份种质中未扩增出条带;共克隆得到120条基因,鉴定得到36种S基因型,并将其中的新基因登录到NCBI中,登录号依次为 OQ124075~OQ124109.
Cloning and sequence analysis of self-incompatibility S-RNase genes from Prunus humilis germplasm resources
The objective of this study was to identify the S genotypes of different Prunus humilis germplasm resources and analyze the sequence characteristics of the Prunus humilis S gene.A total of 70 Prunus humilis germplasm resources were used as test materials.The S gene was amplified through PCR,followed by cloning and sequencing to obtain its gene sequence.Using the BFP93/BFP94-1 primers,two S alleles were identified in each of the 52 germplasm resources.Single S allele was identified in 16 germplasm resources.No bands were amplified in the'3-32-bianhuang'and'10-32'germplasm resources.A total of 120 genes were cloned,and 36 S genotypes were identified.The new genes were registered in NCBI with registration numbers in the following sequence OQ124075~OQ124109.
