贵州火龙果病毒检测及侵染类型分析

为明确贵州省火龙果病毒种类和复合侵染类型,以及主产区病毒发生动态.采集贵州省罗甸县、望谟县、贞丰县、镇宁县火龙果果园的火龙果茎共703份,利用CVX、CVX-NTU、PiVX、ZyVX和SchVX特异性引物对疑似病毒病样品进行RT-PCR检测.结果表明:2021、2022年罗甸县火龙果病毒检出率分别为85.92%、94.90%,望谟县分别为93.48%、93.90%,贞丰县分别为90.99%、99.02%,镇宁县分别为94.52%、94.17%;其中,望谟县在2021年未检出SchVX,2022年则有了较高的检出率(均为复合侵染),表明该病毒在望谟县扩散传播.2021、2022年罗甸县火龙果病毒复合侵染检出率分别为83.10%、90.82%,望谟县分别为93.48%、87.80%,贞丰县分别为90.09%、99.02%,镇宁县分别为93.15%、93.34%.从复合侵染的类型看,2种病毒复合侵染有9种类型,3种病毒复合侵染有10种类型,4种病毒复合侵染有5种类型,另有5种病毒复合侵染的类型.贵州省火龙果的病毒侵染以几种PVX属病毒复合侵染为主,且已在贵州省火龙果主产区大面积发生.
Virus detection and analysis of infection types of pitaya in Guizhou
To clarify the types and mixed infection types of Guizhou pitaya viruses,as well as the dynamics of virus occurrence in the main production areas.A total of 703 pitaya stems were collected from pitaya orchards in Luodian County,Wangmo County,Zhenfeng County,and Zhenning County,Guizhou Province.Suspected viral disease samples were detected by RT-PCR using CVX,CVX-NTU,PiVX,ZyVX,and SchVX specific primers.The detection rates of pitaya virus infection in Luodian in 2021 and 2022 were 85.92%and 94.90%,Wangmo was 93.48%and 93.90%,Zhenfeng was 90.99%and 99.02%,and Zhenning was 94.52%and 94.17%;Among which,in 2021,SchVX was not detected in Wangmo,and in 2022,there was a high detection rate(both of which were mixed infection),indicating the spread of SchVX in Wangmo.In 2021 and 2022,the mixed infection detection rates of pitaya virus were 83.10%and 90.82%in Luodian,93.48%and 87.80%in Wangmo,90.09%and 99.02%in Zhenfeng,and 93.15%and 93.34%in Zhenning.From the detection perspective of mixed infection types,9 types were detected of 2 viruses mixed infection,10 types of 3 viruses mixed infection were detected,5 types of 4 viruses mixed infection were detected,and the type of 5 virus mixed infection.The virus infection of Guizhou pitaya is mainly caused by a combination of several PVX viruses,and has occurred extensively in the main pitaya production areas of Guizhou.
