Breeding of High Erythromycin-producing Strains and Optimization of Fermentation Conditions
In order to select a good strain and optimize fermentation processes for erythromycin fermentation,Saccharopolyspora erythraea SE-2207 strain producing erythromycin was used as the original strain.SE-2207 strain was mutagenized by mutagenesis EMS(ethyl methanesulfonate),UV(ultraviolet)and ARTP(atmospheric pressure room temperature plasmawas),respectively.And the mutagenic effects of the three methods were compared.After mutagenesis,SEY-7 strain with good genetic stability and high erythromycin base tolerance was screened out.Its mycophenolic acid fermentation potency was up to 8368 mg/L,which was 25.5%higher than that of the original strain.Based on screening concentration of N-propanol and soybean oil in the medium,the fermentation processes were optimized and made the fermentation titer of 500 L up to 9155 mg/L.The results were of great significance to improve fermentation production level of erythromycin.
