Establishment and Comparison of Indirect Immunofluorescence Assay for Detection of Marek's Disease Turkey Herpesvirus in Chickens
To establish an indirect immunofluorescence method for detecting Marek's disease turkey herpesvirus(MDV HVT)in chickens,the MDV HVT virus was inoculated into a 96 well cell plate culture(CEF).After 72 hours,chicken anti Marek's disease virus positive serum was used as the primary antibody and rabbit anti chicken fluorescent antibody as the secondary antibody.The reaction conditions were optimized and an indirect immunofluorescence method(IFA)was established.And compared with the classic Malik plaque counting method for virus titer determination,statistical methods were used for comparison,with P less than 0.05 and a correlation coefficient of 0.995,indicating a good correlation between the two methods,.Both methods can be used for titer determination of Malik's disease virus.The IFA method is simpler and faster than the plaque counting method,and can replace the plaque counting method for Preliminary determination of virus content in Malik's disease live vaccines,Accumulate experimental databy adding identification and testing methods for the live Mareks disease vaccine in the 2025 edition of the Chinese Veterinary Pharmacopoeia.
