西南地区市场冰球子分子鉴定

Molecular Identification of Bingqiuzi from Market in Southwest China

利用rDNA ITS和叶绿体trn L-F序列,对17份西南地区冰球子进行分子鉴定.采用改进CTAB法提取独蒜兰叶片基因组DNA,利用通用引物ITS 1和ITS 4以及引物trn L-F对其DNA中rDNA ITS区和叶绿体trnL-F序列进行扩增,对目的片段进行测序并对测序结果进行聚类分析.结果表明,17份西南独蒜兰代表3个种,即云南独蒜兰、独蒜兰和一未定种,药源基本可靠.
The rDNA ITS and chloroplast trnL-F sequences were applied to molecularly identify 17 pleione in southwest China.The genomic DNA of pleione was extract from leaves with the improved CTAB method.Then,the rDNA ITS and chloroplast trnL-F in the DNA were amplified with universal primers ITS 1 and ITS 4 and primer trnL-F separately.The expected fragments were sent to sequece and the results for cluster analysis showed that 17 pleione represent three species,Pleione yunnanensis,P.bulbocodioides and an unknown species,suggesting that the medicinal resouces of Bingqiuzi in Southwest China was basicly reliable.
