Screening and Identification of Silicon Solubilizing Paenibacillus xylanilyticus Strain CBP-2 and Its Effect on Seed Germination
In order to explore and utilize the resources of high-efficiency silicon releasing bacteria and improve the effective silicon content in the soil,silicate selective culture medium was used for the iso-lation and screening of high-efficiency silicon releasing bacteria.The isolated silicon releasing bacteria were classified and identified,and their silicon releasing ability,stress resistance,phosphorus and po-tassium solubilization,iron production carriers,and IAA secretion promoting abilities were measured.The effects of seed soaking and germination inducing experiments on the germination and seedling growth of Chinese cabbage were explored.The results showed that 21 silicon releasing strains were isolated and screened from soil samples,among which strain CBP-2 had an effective silicon concentra-tion of 22.46 mg/L in the fermentation broth using silicon-rich iron tailings as the substrate,which was 2.02 times higher than the non inoculated control.After morphological,physiological and bio-chemical characteristics,and 16S rRNA gene sequence analysis,it was preliminarily identified as Paenibacilli xylanilicus.Silicon releasing bacterium CBP-2 exhibitted good stress resistance and could grow normally at 20 ℃-40 ℃,pH 5-10,and NaCl concentration 0%-7%.The optimal fer-mentation conditions were initial pH 6.0,NaCl concentration 4%,and cultivation at 30 ℃.Strain CBP-2 had certain activation and utilization abilities for calcium phosphate and potassium feldspar,while also producing iron carriers and secreting IAA.This strain had multiple growth promoting func-tions such as silicon release and phosphorus solubilization,as well as good ecological adaptability.It had potential development and application value in soil remediation and agricultural production.
