Transcriptome Analysis of the Effect of UV-B Radiation Treatment on Waxy Maize Seed Germination
In order to understand the molecular mechanism of the effect of medium wave ultraviolet(UV-B)on the activity of waxy maize seeds,waxy maize inbred line N51 seeds were treated with UV-B irradiation at 0 min(control),30 min,1 hours,2 hours,4 hours and 8 hours,and the activity in-dexes and transcriptomics were analyzed.The results showed that there was no significant difference between waxy maize inbred line N51 and the control.In the range of 0 min,30 min,1 hours,2 hours,4 hours and 8 hours,the germination potential,germination index and vitality index all showed a trend of first increasing and then decreasing,and the treatment length reached the highest at 1 hours.According to various vitality indexes,UV-B irradiation at 1 hours had the most significant effect on promoting corn seed germination.According to 0 min(control),1 hours,2 hours 3 treatments,the seed vitality indexes of waxy maize showed a trend of first increasing and then decreasing.Transcrip-tome sequencing analysis screened out 5 differentially expressed genes that were first up-regulated and then down-regulated,and 869 differentially expressed genes that were first down-regulated and then up-regulated.GO annotation classification showed that UV-B treatment mainly affected the differential expression of germination-related phenotypes of waxy maize inbreeding line N51 seeds by affecting th-ylakoids,photosynthetic system light,cooperative use,photosynthetic membrane,photosynthetic antenna protein,metabolic pathway,nitrogen metabolic pathway and other metabolic pathways.The enrichment of KEGG pathway included phenylpropanoid biosynthesis,flavonoid biosynthesis,starch and sucrose metabolism.
