Bioinformatics Analysis of matK Gene in Callicarpa kwangtungensis Chun
The complete nucleotide sequence of Callicarpa kwangtungensis Chun matK gene was obtained by PCR amplification. The open reading frame, amino acid composition, protein transmem-brane structure region, subcellular localization and binding region of matK gene were analyzed by bioinformatics to predict signal peptide, amino acid hydrophilicity/hydrophobicity,amino acid and ad-vanced structure. The UPGMA algorithm was used to construct phylogenetic trees for 13 species of Callicarpa. The results showed that the length of Callicarpa kwangtungensis Chun matK gene was 1 524 bp,encoding 507 amino acids, and there were 10 open reading frames. The matk-encoded ma-ture enzyme K pretein contained 20 amino acids, and the α spiral accounted for 49.11%,β rotation an-gle 4.45%and random curling 28.21%in the secondary structure, which was an unstable hydrophilic protein. No signal peptide,no transmembrane structure, were located to chloroplast suborganelles. The phylogenetic clustering results showed that the relatives of the Callicarpa kwangtungensis Chun were close to Callicarpa bodinieri and Callicarpa mollis
