Research on the Effect of Overexpression Eucommia ulmoides EuDIR1 Gene on Tomato Resistance to Botrytis cinerea
Dirigent(DIR)proteins play an important role in the synthesis of lignin,which improve plant resistance to various pathogens.In this study,based on the Dirigent1 gene of Eucommia ulmoides(EuDIR1)cloned in the laboratory,the EuDIR1 overexpression vector was constructed and transformed into tomato.After obtaining the transgenic plants,the role of EuDIR1 gene in tomato resistance to Botrytis cine-rea was analyzed.The results showed that compared with wild type and unloaded tomato,tomato with over-expression of EuDIR1 gene after inoculation with Botrytis cinerea occurred later,and the lesion diameter was significantly smaller than that of wild type and and plants transferred into the empty vector,indicating that EuDIR1 significantly improved the resistance of tomato plants to Botrytis cinerea.Overexpression of EuDIR1 gene could also increase the protective enzyme activity and the expression of tomato course-related protein(PRs)gene before and after inoculation,thus enhancing the resistance of tomato plants to Botrytis cinerea.In addition,after overexpression of EuDIR1 gene,the lignin content in tomato leaves and stems increased significantly,and the cell wall near the xylem vessel significantly thickened.It was speculated that EuDIR1 gene could enhance tomato resistance to Botrytis cinerea by increasing lignin content in tomato and thickening the cell wall near xylem vessel.This study revealed the function of EuDIR1 gene in tomato resist-ance to Botrytis cinerea,which could provide some theoretical basis for tomato disease resistance breeding.
