不同产地金莲花种质评价及物质形成机制研究

采用显微观察、高效液相色谱法等评价大兴安岭和五台山产地的金莲花种子生物学、显微、化学特性,系统性评价两产地种质差异,并依托转录组学技术进行测序分析种质物质差异形成机制,为金莲花种质筛选及植物的品种选育提供重要参考.结果表明,大兴安岭金莲花种子含水率、吸水率、生活力等生物学特性优于五台山产地.经HPLC法测定大兴安岭荭草苷和牡荆素含量分别为0.014 3~0.014 6 mg/g、0.840~0.868 mg/g.转录组学测序共拼接得到Unigene 32 959条,获得差异基因3 165个.大兴安岭金莲花种质优势可能与调控S-腺苷-L-蛋氨酸3'-羟基-N-甲基氯嘌呤4'-O-甲基转移酶、乙醛脱氢酶(NAD(P)(+))ald5、查尔酮合成酶2、植物过氧化物酶等关键酶的基因参与苯丙烷类物质生物合成、异喹啉生物碱生物合成、类黄酮生物合成等密切相关.
Study on Germplasm Evaluation and Formation Mechanisms of Trollius chinensis Bunge from Different Producing Areas
Microscopic observation and high-performance liquid chromatography were used to analyze the biological,microscopic,and chemical properties of seeds from Daxinganling and Shanxi Province,and tran-scriptome sequencing was used to perform the sequencing study on substance formation difference mechanisms of germplasms,in order to provide an essential reference for germplasm screening and plant variety selection.The results showed that the biological characteristics like seed moisture,water absorption and viability of the Trollius chinensis Bunge seeds from Daxinganling were generally better than those from Shanxi Province.The HPLC method was used to determine the contents of orientin and vitexin in the Daxinganling.The contents of orientin and vitexin in Daxinganling were 0.014 3-0.014 6 mg/g and 0.840-0.868 mg/g,respectively.Tran-scriptome sequencing was performed on the seeds of the two regions,and a total of 32 959 Unigenes were assembled.3 165 genes with differential expression were found in total,and S-adenosyl-L-methionine:3'-hy-droxy-N-methylcoclaurine 4'-O-methyltransferase,aldehyde dehydrogenase(NAD(P)(+))ald5,chalcone synthase 2,plant peroxidase,and other key enzyme genes could be closely related to phenylpropanoid biosyn-thesis,isoquinoline alkaloid biosynthesis,and flavonoid biosynthesis.
