Establishment of Rapid Propagation System in Vitro of Verbena bonariensis
This study used potted seedlings of Verbena bonariensis as experimental materials to explore its rapid propagation system.The results showed that a 5-6 cm stem segment with axillary buds was designed with a combination of 75%alcohol and 20%NaClO.It was found that the best effect was achieved under the conditions of 75%alcohol disinfection for 30 seconds and 20%NaClO disinfection for 8 minutes.After 7 days,the pollution rate was 12.78%,the mortality rate was 4.44%,and the survival rate was 82.78%.The 3-4 cm buds were cultured in MS medium with different concentra-tions of 6-BA and NAA for proliferation induction.It was found that when the medium was MS+1.5 mg/L 6-BA+0.10 mg/L NAA,the induction rate was 100%after 30 days,and the proliferation coef-ficient reached 4.77,resulting in a large number of clustered buds.3-4 cm clusters of buds were cut into single buds and culture them on MS+0.10 mg/L NAA medium.After 15 days,the height differ-ence of the plant was 4.64 cm,and the seedlings were robust and without withering.The rootless seedlings of Verbena bonariensis with good growth and a height of about 5 cm were placed in a culture medium to explore the concentration of IBA and NAA hormones.Under the conditions of MS+0.10 mg/L IBA+0.05 mg/L NAA,the rooting rate was 100%after 15 days,with an average rooting number of 40.43 and an average root length of 3.27 cm.After 45 days,the average root length was 14.33 cm.Further replacing agar with vermiculite as the substrate,the rooting rate was 100%after 15 days,and the average root length could reach 4.50 cm.After 45 days,the average root length could reach 42.33 cm,significantly greater than the average root length in the agar substrate.When the root length and number of Verbena bonariensis plants in the root culture medium were suitable,sterile seedlings with similar growth were selected for subsequent seedling cultivation and transplantation.It was found that after 4 days of seedling cultivation under natural light conditions,they were transplan-ted onto a substrate of Vvermiculite∶Vnutrient soil=1∶2,and after 30 days,the survival rate was 93.33%.
