Establishment of Tissue Culture and Regeneration System of Abelmoschus esculentus L.
In this experiment,the leaves and stem segments of Abelmoschus esculentus L.were used as explants,and the effects of different hormone combinations on callus induction,callus proliferation,adventi-tious bud differentiation and rooting were studied by tissue culture.The best medium formula suitable for each stage was selected,and the disinfection effect of different disinfectants was briefly explored.The results showed that the best disinfection method was 75%C2H5OH soaking for 2 minutes+0.15%HgCl2 soaking for 20 minutes.The best medium for callus induction was MS+2.0 mg/L 6-BA,and the induction rate of stem segments with nodes reached 98%.The optimal medium for proliferation was MS+2.0 mg/L 6-BA+0.2 mg/L NAA,and the proliferation coefficient was 4.The best differentiation medium was MS+2.0 mg/L ZT+0.15 mg/L NAA,and the differentiation rate was 93%.The best rooting medium:MS+0.1 mg/L NAA,the root quality was good,the main root was strong,and the root system was developed.This experiment improved the process of tissue culture and regeneration system establishment of Abelmoschus esculentus L.,and contributed reference value for the protection,utilization,genetic transformation of its germplasm resources and tissue culture research of other plants.
