Establishing and Optimizing the Tissue Culture and Rapid Propagation System of Pueraria montana var.thomsonii Gan'ge No.2
In order to accelerate the breeding and promotion of the high-quality seedlings of Gan'ge No.2,a local variety of Pueraria montana var.thomsonii,Hengfeng County,this study used stem segment with buds as materials to discuss the establishment of aseptic system,induction and prolifer-ation of advective buds,rooting of test-tube seedlings,acclimation and transplanting,and established and optimized the tissue culture rapid propagation system of Pueraria montana var.thomsonii Gan'ge No.2.The results showed that the optimal medium was MS+6.5 g/L agar+30 g/L sucrose+1.5 mg/L 6-BA+1 mg/L NAA,with an average induction rate of 80.55%.The optimal medium for subculture was MS+6.5 g/L agar 30 g/L sucrose+1 mg/L 6-BA+0.5 mg/L NAA,with an average induction rate of 91.66%.The optimal medium for rooting was MS+6.5 g/L agar+30 g/L sucrose+0.5 mg/L NAA.The average induction rate was 84.38%.The transplanting substrate formulation was Vpeat soil∶Vvermiculite∶Vperlite=3∶2∶1,and the survival rate of the seedlings was 100%.
