New Haplotype of Salt Tolerance Gene TaHKT1 and Evaluation of Salt Tolerance in Wheat
In this study,under the 0.5%salt stress treatment,275 wheat varieties(lines)were identi-fied for salt tolerance during the germination stage,and relative indicators such as germination rate,shoot length,root length,root number,shoot fresh weight,root fresh weight,and root crown ratio were measured.The salt tolerance of 7 indicators was simplified into 3 principal components using principal component analysis,with a cumulative contribution rate of 74.35%.Through the analysis of principal component contribution rate and membership function,the 3 principal component values were further reduced to D-value of comprehensive evaluation index.According to D-value and cluster analysis,275 wheat varieties(lines)were divided into 5 classes of salt resistance,including high salt tolerance,salt tolerance,medium salt tolerance,salt sensitivity,and high salt sensitivity.Eleven,one hundred and seven,and sixty wheat varieties(lines)with high salt tolerance,medium salt toler-ance,and salt tolerance above were selected,accounting for 64.7%of the total.The TaHKT1 gene sequence analysis of 13 wheat varieties(lines)revealed that the new T∶T haplotype was related to salt tolerance.275 wheat varieties(lines)were tested by KASP markers,98 of which were the new haplo-type,accounting for 35.64%.The proportion of new haplotype wheat varieties(lines)in different provinces were different.The correlation analysis between the T∶T genotype of wheat varieties(lines)and the salt tolerance D-value showed that the new haplotype of TaHKT1 gene was significantly dif-ferent from the salt tolerance of wheat at the bud stage(p<0.01).
