基于SSR分子标记的紫苏种质资源遗传多样性及群体结构分析

为了明确不同紫苏种质的遗传基础和群体的亲缘关系,本研究利用19对SSR分子标记对不同地理来源的141份紫苏资源进行遗传多样性和群体结构分析.结果表明,19对SSR分子标记扩增出80条多态性条带,平均每对引物4.26条,多态性指数(PIC)变幅在0.331 1~0.845 7之间,平均为0.620 6,多态性条带占比85.10%.利用UPGMA法对141份紫苏进行分子聚类,遗传相似系数(GS)介于0.478 7~0.948 9之间,平均为0.709 9,在GS=0.696处将供试材料分为三类.通过Structure群体结构分析,将供试材料分为三大类,其中68.8%的紫苏种质资源遗传背景比较单一,31.2%的资源拥有混合血统,其遗传背景较为复杂.
Genetic Diversity and Population Structure Analysis of the Perilla frutescens L.Germplasm Resources Based on SSR Markers
In order to clarify the genetic basis of different Perilla frutescens L.germplasm and popula-tion relationship,19 pairs of SSR molecular markers were used to analyze the genetic diversity and population structure of 141 Perilla frutescens L.resources from different geographical sources.The results showed that 80 polymorphic bands were amplified from 19 pairs of SSR markers,with an av-erage of 4.26 bands per primer pair.The polymorphism index(PIC)ranged from 0.331 1 to 0.845 7,with an average of 0.620 6 bands,accounting for 85.10%.Molecular clustering of 141 Perilla was per-formed by UPGMA method,and the genetic similarity coefficient(GS)ranged from 0.478 7 to 0.948 9,with an average of 0.709 9.The materials were divided into three classes at GS=0.696.Through the analysis of Structure and population structure,the tested materials were divided into 3 groups.Among them,68.8%of Perilla frutescens L.germplasm resources had a single genetic back-ground,31.2%had mixed ancestry,and their genetic background was more complex.
