Genetic Diversity Analysis of Cultivated Auricularia heimuer Based on ISSR and TRAP Molecular Markers
In order to explore the genetic diversity and genetic relationship of cultivated Auricularia heimuer strains in northeast China,genetic diversity of 50 cultivated Auricularia heimuer strains was analyzed by ISSR and TRAP molecular markers.The DNA of cultivated Auricularia heimuer was amplified by 22 highly polymorphic and high-resolution ISSR and TRAP labeled primers.The results showed that the polymorphisms of ISSR and TRAP markers were high,accounting for 89.58%and 84.71%,respectively,with genetic similarity coefficients ranging from 0.53 to 1.00.The strains with similarity coefficient greater than 0.96 accounted for 44.0%in ISSR labeled cluster plots,and 54.0%in TRAP labeled cluster plots.The STRUCTURE clustering results showed that the 50 strains could be divided into four categories.The results of genetic diversity analysis showed that the genetic diver-sity of Auricularia heimuer cultivated strains in northeast China was mainly from inter-population,and the genetic diversity of Auricularia heimuer cultivated strains was not rich,and the homogeneity of production strain resources was prominent.
