不同培养基对固沙先锋植物羽毛针禾种子萌发的影响

为探索固沙先锋植物羽毛针禾种子萌发的最适培养基条件,以羽毛针禾种子为材料,利用MS、N6、1/2MS、琼脂和White等5种培养基进行羽毛针禾种子萌发实验.在筛选出高效(琼脂培养基)和低效(White)培养基后,对种子进行可溶性糖含量、可溶性蛋白含量、α-淀粉酶活性以及赤霉素、脱落酸、乙烯利、茉莉酸和生长素等萌发相关指标的检测.结果表明,琼脂培养基中种子萌发率最高,种子萌发前可溶性蛋白含量和α-淀粉酶活性持续升高,可溶性糖含量基本保持稳定,相关植物激素赤霉素、脱落酸、乙烯利、茉莉酸和生长素含量处于持续上升的状态.White培养基中萌发率最低,种子萌发前α-淀粉酶活性持续升高,可溶性蛋白含量显著下降,赤霉素浓度显著低于琼脂糖培养基,脱落酸的含量高于琼脂糖培养基.研究结果表明,琼脂培养基为5种羽毛针禾种子萌发的最高效培养基.
The Effect of Different Culture Media on the Germination of the Sand Fixing Pioneer Plant Stipagrostis pennata Seeds
In order to explore the optimum medium conditions for seed germination of Stipagrostis pennata,a pioneer plant of sand fixation,MS,N6,1/2 MS,AGAR and White media were used for seed germination experiments.After high efficiency(AGAR medium)and low efficiency(White)medium were selected,the seeds were tested for soluble sugar,soluble protein,α-amylase,germinal,ababolic acid,ethethylene,jasmonic acid and growth hormone.The results showed that the seed ger-mination rate was the highest in AGAR medium.Before germination,soluble protein content andα-amylase activity continued to increase,soluble sugar content remained stable,and the contents of related plant hormones gibberellin,ababolic acid,ethethylene,jasmonic acid and auxin continued to increase.Before germination,α-amylase activity continued to increase,soluble protein content decreased significantly,gibberellin concentration was significantly lower than agarose medium,and abscisic acid content was higher than agarose medium.The results showed that AGAR medium was the most efficient medium for seed germination of 5 species of Stipagrostis pennata.
