基于非靶向代谢组学研究天麻块茎抽薹前后的差异代谢产物

本研究采用代谢组学方法鉴定并分析了天麻抽薹前后的小分子代谢物质,以了解天麻块茎从休眠期到抽薹期的发育过程.通过LC-MS/MS检测、主成分分析、正交偏最小二乘判别及通路分析,结果表明,抽薹前后共鉴定出65个显著差异代谢物,分别为氨基酸及其衍生物、有机酸及其衍生物、脂肪酸及其衍生物、生物碱、芳香族、核苷/氨基酸、核苷、核苷酸、硫脲、糖类、萜类、维生素、甾体、甾体皂苷等14类化合物,其中上调代谢物主要是氨基酸及其衍生物类型化合物、生物碱等,下调代谢物主要是脂肪酸及其衍生物和有机酸及衍生物.KEGG分析显示,差异代谢物主要富集到苯并恶嗪类生物合成、精氨酸和脯氨酸代谢、玉米素的生物合成等相关的通路上.
Study on Differential Metabolites of Gastrodia elata Tuber before and after Bolting Based on Non-targeted Metabolomics
In this study,metabolomics was used to identify and analyze the small molecule metabolites of Gastrodia elata before and after bolting,in order to understand the development process of Gastro-dia elata tuber from the dormant stage to the bolting stage.Through LC-MS/MS detection,principal component,orthogonal partial least squares discrimination and path analysis,the results showed that 65 metabolites with significant differences were identified before and after bolting,including amino acids and their derivatives,organic acids and their derivatives,fatty acids and their derivatives,alka-loids,aromatics,nucleosides/amino acids,nucleosides,nucleotides,thiourea,sugars,terpenes,vitamins,steroids,steroid saponins and other 14 types of compounds,respectively,among which the up-regulated metabolites were mainly amino acids and their derivatives,alkaloids,etc.,and the down-regulated metabolites were mainly were mainly fatty acids and their derivatives and organic acids and derivatives.KEGG analysis showed that the differential metabolites were mainly concentrat-ed in the pathways related to benzoxazine biosynthesis,arginine and proline metabolism,and zeatin biosynthesis.
