Cloning and Expression Characteristics of RcNAC91 Gene in Ricinus communis
In order to investigate the role of NAC family genes in the anti-abiotic stress response in Ricinus communis,an upregulated RcNAC91 gene(GenBank No.OR756194)was cloned from the salt stress transcriptomic data of Ricinus communis by RT-qPCR bioinformatics analysis.The tissue expression characteristics and response to stress were detected by RT-qPCR.The results showed that the RcNAC91 gene was located on chronosome No.1,with the cDNA sequence of 1 911 bp,including 1 755 bp coding region and 284 amino acids were inferred,which was a hydrophilic protein with a transmembrane domain.Subcellular localization showed that the RcNAC91 protein was localized in the nucleus.The phylogenetic tree results showed that RcNAC91 had the closest relationship with NAC protein of Pseudognaphalium affine.The RcNAC91 promoter region had multiple stress response and hormone-induced elements.RT-qPCR results showed that the expression of RcNAC91 gene was the highest in the cotyledon,and the expression of RcNAC91 gene was activated by drought,salt,low temperature and abscisic acid stress,indicating that RCNAC91 gene could actively participate in the abiotic stress response of Ricinus communis.
