基于转录组测序对黄秋葵籽内参基因的发掘与验证

为获得黄秋葵籽不同发育时期均稳定表达的内参基因,本研究对授粉后3 d、6 d、15 d、25d和40 d的黄秋葵籽粒进行转录组测序,基于黄秋葵全基因组注释信息和黄秋葵籽转录组测序数据,筛选出6个新的候选内参基因ZCCHC11、SEU、RPL11、FKBP62、MBR2L、BOB1,与文献报道的 6 个常用持家基因 GAPDH、EF1-α、TUA5、YLS8、ACT2、eIF4A的稳定性进行对比.采用qRT-PCR检测上述基因在不同发育时期黄秋葵籽中的表达水平,运用geNorm、NormFinder、BestKeeper和RefFinder综合评价其表达稳定性,然后利用淀粉合成酶基因AeAGPL2对筛选出的内参基因进行验证.结果表明,ZCCHC11L和SEU的稳定性最佳,TUA5稳定性最差;新发掘的内参基因ZCCHC11L、SEU及其ZCCHC11L+SEU组合可作为黄秋葵籽不同发育时期的内参基因.
Excavation and Verification on Internal Reference Genes in Abelmoschus esculentus Seeds Based on Transcriptome Sequencing
To identify stably expressed reference genes in Abelmoschus esculentus seeds at different developmental stages,transcriptomic sequencing was conducted on okra seeds at 3 days,6 days,15 days,25 days and 40 days post-pollination.Based on the whole genome annotation of Abelmoschus es-culentus and transcriptome sequencing data of Abelmoschus esculentus seeds,six new candidate internal reference genes ZCCHC11L,SEU,RPL11,FKBP62,MBR2L and BOB1 were compared for stability with six commonly used housekeeping genes GAPDH,EF1-α,TUA5,YLS8,ACT2 and eIF4A reported in literature.qRT-PCR was employed to assess gene expression in Abelmoschus esculentus seeds at various growth stages.The expression stability of selected internal reference factors was comprehensively evaluated using geNorm,NormFinder,BestKeeper and RefFinder.Subsequently,the chosen internal reference genes were validated against the starch synthetase gene AeAGPL2.Results indicated that ZCCHC11L and SEU exhibited optimal stability while TUA5 displayed the least stable expression.The newly identified internal reference genes ZCCHC11L and SEU as well as their combination ZCCHC11L+SEU could be used as internal reference genes at dif-ferent developmental stages of Abelmoschus esculentus seeds.
