Screening of the Methods for Detecting the Pollen Viability of Paeonia ludlowii
In order to select a suitable method for the detection of Paeonia ludlowii pollen viability,using Paeonia ludlowii pollen as research object,the pollen viability was determined by culturing in vitro and stai-ning methods(TTC,iodine-potassium iodide and carmine acetate).The results showed that the effect of four factors on pollen germinatin rate in vitro of Paeonia ludlowii from strong to weak was temperature,sucrose concentration,boric acid concentration,calcium chloride concentration,of which temperture and sucrose con-centration had the most significant effect on pollen germination rate in vitro.Under the existing factors,1%AGAR+150 g/L sucrose+20 mg/L calcium chloride+50 mg/L boric acid at constant temperature of 25 ℃ culturing for 1.5 hours was the best condition for in vitro culture of Paeonia ludlowii pollen.The comparison of four methods showed that in vitro pollen culture could be used for rapid and accurate determination of pollen viability,and iodine-potassium iodide staining could be used for rapid detection of pollen viability.TTC staining and acetic acid magenta staining rules were not applicable.
