基于ISSR分子标记的苗药朱砂根遗传多样性研究

为了探究不同居群朱砂根遗传多样性水平及亲缘关系,以24份不同居群朱砂根为材料,采用ISSR分子标记技术对其进行筛选分析,结果从100条引物中筛选到14条扩增效果好、多态性高的ISSR引物,对24个朱砂根居群进行遗传多样性分析,共检测到75条扩增位点,平均每条引物扩增5.4个位点,74个为多态性位点,多态性比率(PPB)为98.67%;其中,UBC841扩增位点最多(8个),UBC820、UBC821、UBC844、UBC887扩增位点最少(4个).观测等位基因数为2,有效等位基因数为1.086 8~2.000 0,Nei's基因多样度为0.079 9~0.500 0,Shannons指数最大,为0.693 1;遗传距离与地理距离无显著相关性.研究表明,朱砂根种质资源遗传多样性高,ISSR分子标记可用于朱砂根亲缘鉴定.
Study on Genetic Diversity of Miao Medicine Ardisia crenata Root Based on ISSR Molecular Markers
In order to explore the genetic diversity level and relationship of different populations of Ardisia crenata,24 different populations of Ardisia crenata were selected and analyzed by ISSR molecular marker technology.The results showed that 14 primers with good amplification effect and high polymorphism were selected from 100 primers,and genetic diversity was detected in 24 popula-tions of Ardisia crenata.There were 75 amplification sites detected,with an average of 5.4 amplifica-tion sites per primer,74 polymorphic sites,and polymorphism ratio(PPB)was 98.67%.UBC841 had the most amplification sites(8),and UBC820,UBC821,UBC844 and UBC887 had the least amplifi-cation sites(4).The observed number of alleles was 2,the effective number of alleles was 1.086 8-2.000 0,the gene diversity of Nei's was 0.079 9-0.500 0,and the Shannon's index was the largest(0.693 1).The correlation analysis between genetic distance and geographical distance were found no significant correlation.Study showed that the genetic diversity of Ardisia crenata germplasm resources was high,and ISSR molecular markers could be used to identify the relatives of Ardisia crenata.
