摘要
为了揭示种子活力与TTC染色之间的定量关系,本研究以水稻种子CY-150为试验材料,经人工加速老化处理和TTC染色后,利用体视镜获取种子胚部纵切面的RGB图像,并用农业农村部植物新品种测试(昆明)分中心建立的图像分析软件分析图像的CIELab值,计算单位面积染色强度.结果表明,发芽率、TTC染色率和活力指数之间呈显著正相关.其中,发芽率和活力指数、发芽率和染色率、染色率和活力指数之间的相关系数分别为0.997 6、0.971 8和0.963 1.此外,CIELab值与发芽率、染色率和活力指数之间显著相关,其中,L*和a*分别呈负相关和正相关,相关系数分别为-0.974 9、-0.976 1、-0.972 1、0.973 7、0.991 5和0.967 8.利用种子活力指数与CIELab值建立了种子活力方程.通过随机选取4个水稻品种对方程进行验证,结果显示,4个品种的活力指数真实值略高于预测值.研究表明,TTC染色不仅可用于水稻种子活力快速定性分析,而且通过种胚RGB图像可以实现种子活力的定量分析,为进一步普及水稻种子活力快速检测提供参考.
Abstract
In order to reveal the quantitative relationship between seed vitality and TTC staining,this study took rice(Oryza sativa L.)seed CY-150 as the experimental material.After artificial acceler-ated aging treatment and TTC staining,stereoscopic RGB images of the longitudinal section of seed embryo were obtained,and the CIELab values of the images were analyzed by the image analysis soft-ware established by the Plant New Variety Testing(Kunming)Sub-center of the Ministry of Agricul-ture and Rural Affairs,and calculating the staining intensity per unit.The results showed that there was a significant positive correlation between germination rate,TTC staining rate and vigor index.Among them,the correlations between germination rate and vitality index,germination rate and stai-ning rate,staining rate and vitality index were 0.997 6,0.971 8 and 0.963 1.In addition,CIELab value was significantly correlated with germination rate,staining rate and vitality index,in which L*and a*were negatively correlated and positively correlated,and Pearson correlation coefficients were-0.974 9,-0.976 1,-0.972 1,0.973 7,0.991 5 and 0.967 8,respectively.At the same time,the seed vigor equation was established using seed vigor index and CIELab value.Four rice varieties were randomly selected to verify the equation,and the results showed that the real values of the vigour index of the four varieties were slightly higher than the predicted values.The results showed that TTC staining could not only be used for rapid qualitative analysis of rice seed viability,but also could be used for quantitative analysis of seed viability through seed embryo RGB images,which provided a reference for further popularization of rapid detection of rice seed viability.
