地黄与天目地黄种间杂种倍性及SSR分子标记鉴定研究

为鉴定地黄与天目地黄的杂交后代(粉花地黄)染色体倍性及杂种真实性,本研究采用流式细胞术开展了染色体倍性研究,采用SSR分子标记技术鉴定杂种真实性.结果表明,粉花地黄为四倍体,与母本地黄一致,父本天目地黄为二倍体.通过PCR技术从45对引物中筛选出11对SSR引物用于杂种真实性鉴定,11对引物共扩增出不同条带总数为45,多态性条带数为40,多态性比率达88.89%,除引物CPSSR_225外,从其余每对引物扩增出的图谱中可见,粉花地黄出现的基因峰值均与父本及母本的部分峰值相同,在某个位点中出现4个峰值,各自有2个分别与母本及父本一致.研究表明,粉花地黄为地黄与天目地黄的真杂种.
Study on the Identification about Ploidy and SSR Molecular Marker of Interspecific Hybrids between Rehmannia glutinosa and Rehmannia chingii
In order to identify the chromosome ploidy and hybrid authenticity of the hybrid offspring of Rehmannia glutinosa and Rehmannia chingii,the chromosome ploidy was studied by flow cytome-try,and the hybrid authenticity was identified by SSR molecular marker.The results showed that Rehmannia glutinosa'Fenhua'was tetraploid,consistent with the maternal Rehmannia glutinosa,and the paternal Rehmannia chingii was diploid.By PCR,11 pairs of SSR primers were selected from 45 pairs of primers for the identification of hybrid authenticity.The total number of different bands was 45,the number of polymorphic bands was 40,and the polymorphism ratio was 88.89%.Primers were removed except CPSSR_225,and it was found from the amplified maps of other primers that the peak value of genes in Rehmannia glutinosa'Fenhua'was the same as the partial peak value of paternal and maternal genes,and there were 4 peaks in one locus,2 of which were consistent with the maternal and paternal genes respectively.The results showed that Rehmannia glutinosa'Fenhua'was the true hybrid of Rehmannia glutinosa and Rehmannia chingii.
