秋水仙素处理对牛至种子萌发和幼苗生长的影响

为探究秋水仙素处理对牛至种子萌发及幼苗生长的影响,本试验以牛至种子为材料,采用不同浓度(0(ck)、0.01%、0.05%、0.10%、0.50%)秋水仙素处理牛至种子,分别测定牛至种子萌发期、幼苗生长期的生理生化指标.结果表明,随着秋水仙素浓度升高,牛至种子的发芽率、发芽势呈先升后降的趋势,在0.05%秋水仙素处理下,牛至种子的发芽势和发芽率达到最高值,与对照差异显著,是对照的1.3倍;0.50%秋水仙素处理对种子萌发有抑制作用.随着秋水仙素浓度的增加,牛至幼苗根长逐渐降低,根粗和叶厚逐渐增加.在0.01%的秋水仙素处理下,牛至幼苗株高、叶长、叶宽、叶面积达到最大值,随着秋水仙素浓度的增加,牛至幼苗株高、叶宽等生长指标降低,POD活性呈先增加后降低的趋势,可溶性蛋白和可溶性糖含量高于ck,均在0.01%时达到最高值.对牛至种子测定指标进行隶属函数分析得出,0.01%秋水仙素处理下的平均隶属函数值排名第一.综合分析得出,0.01%秋水仙素浸种24 h是促进牛至种子萌发和幼苗生长的最佳处理.
The Effect of Colchicine Treatment on the Germination and Seedling Growth of Origanum vulgare L.
In order to explore the effects of colchicine treatment on Origanum vulgare L.seed germination and seedling growth,using Origanum vulgare L.seeds as research materials,Origanum vulgare L.seeds were treated with different concentrations(0(ck),0.01%,0.05%,0.10%,0.50%),and the physiological and biochemical indexes of Origanum vulgare L.seeds at germination stage and seedling growth stage were deter-mined respectively.The results showed that with the increase of colchicine concentration,the germination rate and germination potential of Origanum vulgare L.seeds increased first and then decreased.Under the treat-ment of 0.05%colchicine,the germination potential and germination rate of Origanum vulgare L.seeds reached the highest value,which was 1.3 times higher than that of the control.The seed germination was inhibited by 0.50%colchicine treatment.With the increase of colchicine concentration,the root length of Origanum vulgare L.seedlings gradually decreased,and the root thickness and leaf thickness gradually increased.Under 0.01%colchicine treatment,plant height,leaf length,leaf width and leaf area of Origanum vulgare L.seedlings reached the maximum value.With the increase of colchicine concentration,plant height and leaf width of Origanum vulgare L.seedlings decreased,POD activity increased first and then decreased,and soluble protein and soluble sugar contents were higher than ck,reaching the highest value at 0.01%.According to the membership function analysis of Origanum vulgare L.seed determination indics,the aver-age membership function value under 0.01%colchicine ranked first.The results showed that soaking 0.01%colchicine for 24 hours was the best treatment to promote seed germination and seedling growth of Origanum vulgare L..
