金花菜愈伤组织诱导及其试管苗生长发育研究

为了探究金花菜愈伤组织诱导及其试管苗生长情况,本试验以3个金花菜品种(上海金花菜、大叶金花菜和中叶金花菜)为材料,对其无菌体系、愈伤组织的诱导和增殖试管苗的生长发育及其相关生理生化指标进行研究.结果表明,3个金花菜品种无菌体系建立的较佳消毒方式为:75%酒精1 min+0.1%升汞10~15 min;光照条件下,MS+KT 0.5 mg/L+2,4-D2 mg/L有利于提高胚轴愈伤组织诱导过程中可溶性蛋白和可溶性糖含量,提高过氧化物酶(POD)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性,降低胚轴愈伤组织诱导过程中的丙二醛(MDA)含量,是3个金花菜品种愈伤组织诱导的较佳培养基;黑暗条件下,MS+6-BA 2 mg/L+激动素(KT)2 mg/L+2,4-D 0.5 mg/L+NAA 0.5 mg/L有利于提高愈伤组织增殖过程中可溶性蛋白和可溶性糖含量,及其POD、SOD、CAT活性,降低愈伤组织增殖过程中的MDA含量,是3个金花菜品种愈伤组织增殖的较佳培养基;光照条件下,MS+6-BA 1 mg/L+NAA 0.2 mg/L+壳寡糖(COS)0.5 mg/L和MS+KT 1 mg/L+NAA0.2 mg/L+COS0.5 mg/L有利于提高试管苗的可溶性蛋白和可溶性糖含量,及其POD、SOD、CAT活性,降低试管苗的MDA含量,是3个金花菜品种试管苗生长发育的较佳培养基.
Study on Callus Induction and Its Growth and Development of in Vitro Plantlets of Medicago polymorpha L.
In order to investigate the callus induction and the growth of in vitro seedlings of 3 varieties of Medicago polymorpha L.(Shanghai,Daye,Zhongye),the sterile system,callus induction and proliferation of in vitro seedlings and related physiological and biochemical indexes were studied.The results showed that the best disinfection method for the three varieties of Medicago polymorpha L.aseptic system was 75%alcohol for 1 minutes+0.1%mercury bichloride from 10 to 15 minutes.Under light conditions,MS+KT 0.5 mg/L+2,4-D 2 mg/L was beneficial to increase the contents of soluble protein and soluble sugar,as well as the activities of POD,SOD and CAT in the induction process of cotyl callus.The content of MDA in the callus induction process of the cotyl decreased,which was the best medium for callus induction of 3 varieties of Medicago polymorpha L..Under dark conditions,MS+6-BA 2 mg/L+KT 2 mg/L+2,4-D 0.5 mg/L+NAA 0.5 mg/L could improve the contents of soluble protein and soluble sugar,and the activities of POD,SOD and CAT during callus proliferation,and reduce the MDA during callus proliferation,which was the best medium for the proliferation of callus of 3 varieties of Medicago polymorpha L..Under the light condition,MS+6-BA 1 mg/L+NAA 0.2 mg/L+chito-oligosaccharides(COS)0.5 mg/L and MS+KT 1 mg/L+NAA 0.2 mg/L+COS 0.5 mg/L could improve the contents of soluble protein and soluble sugar of the seedlings,of which the POD,SOD and CAT activities decreased the content of MDA in test tube seedlings,and it was a better medium for the growth and development of three varieties of Medicago polymorpha L.seedlings.
