Analysis of SSR Sequence Characteristics for Pyrostegia venusta Based on Transcriptome Data
In this study,the transcriptome SSR loci of Pyrostegia venusta were excavated in order to understand the distribution characteristics of SSR loci and lay a foundation for the development of SSR markers.The results showed that 85 699 Unigenes were obtained from the transcriptome sequencing data of Pyrostegia venusta,and a total of 22 864 SSR loci were found on 18 282 Unigenes.The occurrence frequency of SSR loci was 26.68%,1 SSR locus every 4.38 kb with an average length of 13.89 bp.The main types of repeats for SSR loci of Pyrostegia venusta were single nucleotide repeats(13 780,accounting for 60.27%),followed by dinucleotide repeats(6 141,accounting for 26.86%)and trinucleotide repeats(2 656,accounting for 11.62%).Among the 72 repetitive primitives,the single nucleotide A/T was the most dominant(12 731,accounting for 55.68%),followed by dinucle-otide dominant AG/CT(2 522,accounting for 11.03%)and trinucleotide dominant AAG/CTT(745,accounting for 3.26%).The number of repeats for SSR loci of Pyrostegia venusta was mainly from 5 to 15 times,of which 6 nucleotide repeats decreased with the increase of the number of primitive repeats.The SSR loci mainly focused from 10 to 18 bp,within which the number of SSR loci in this range accounted for 81.87%of the total number of SSR loci.The number of SSR loci with high poly-morphism potential(length>20 bp)was 640,accounting for 10.17%.In conclusion,the frequency of SSR loci of Pyrostegia venusta was high,the SSR loci were abundant,and the polymorphism potential was high.
