多倍体芦竹松散型胚性愈伤组织的诱导

为了建立高效的芦竹体胚发生体系,本试验以带节间的多倍体芦竹组培苗幼嫩茎段为外植体,采用含有不同激素种类和浓度的培养基,在多次继代选择的条件下进行松散型胚性愈伤组织的诱导试验.结果表明,MS+0.25 mg/L 6-BA+1 mg/L 2,4-D为诱导芦竹松散型愈伤组织的最佳培养基,具有愈伤组织诱导速度较快,结构松散,质地较软,呈白绿色的特点;芦竹松散型胚性愈伤组织诱导的最佳培养基为1/2 MS+0.50 mg/L 6-BA+0.50 mg/L 2,4-D,通过电镜观察到愈伤组织全部呈胚性状态,结构松散、质地较硬、生长快速.在无激素的MS培养基上,将诱导出的芦竹松散型胚性愈伤组织进行转移,获得发育状态良好的胚状体需要经过5次继代培养.
Induction of Loose Embryogenic Callus of Polyploid Arundo donax L.
In order to establish an efficient system of somatic embryogenesis of Arundo donax L.,the young stem segments of polyploid Arundo donax L.tissue culture seedlings with interganglionic seg-ments were selected as explants,and the loose embryogenic callus was induced by medium containing different hormone types and concentrations under the conditions of multiple subgenerations.The results showed that MS+0.25 mg/L 6-BA+1 mg/L 2,4-D was the best medium for inducing loose callus of Arundo donax L..It had the characteristics of fast callus induction,loose structure,soft texture and white and green color.The best medium for inducing loose embryogenic callus was 1/2 MS+0.50 mg/L 6-BA+0.50 mg/L 2,4-D.The embryogenic callus showed loose structure,hard tex-ture and fast growth by electron microscopy.The induced loose embryoid callus was transferred on hormone-free MS medium,and 5 subcultures were needed to obtain well-developed embryoid.
