Fingerprints Construction and Genetic Diversity Analysis of Vitis vinifera Germplasm Resources Based on SSR Markers
Taking 119 germplasm resources of wine grape and rootstocks as experimental materials,DNA fingerprinting was constructed and genetic diversity was analyzed using SSR molecular marker technology.The results showed that a total of 165 alleles were amplified by 9 pairs of primers,with the number of alleles amplified by each primer ranging from 15 to 21,and the variation range of poly-morphism information was 0.81 to 0.90,with an average of 0.86.Each pair of primers could distinguish 14 to 35 varieties.Except for different strains of the same variety that could not be distin-guished by using 9 pairs of primers,the remaining germplasms could be completely distinguished by using 3 pairs of primers of VVMD28+VVMD7+VVMD27.Eventually,119 fingerprint maps of grape germplasms were constructed using 3 pairs of SSR primers.UPGMA cluster analysis indicated that at the genetic similarity coefficient of 0.827,119 germplasms could be classified into three major categories:The first major category included 99 grape germplasms such as Vitis vinifera species,hybrids between Vitis vinifera and Vitis labrusca,Vitis quinquangularis,Vitis amurensis and its derived wine grape varieties;the second major category consisted of 12 rootstocks,including Vitis ri-paria species,hybrids with Vitis riparia as the female parent,and hybrids with Vitis amurensis as the female parent and Vitis riparia as the male parent;the third major category contained 8 root-stocks with Vitis berlandieri as the female parent.The clustering results basically reflected the genet-ic relationships among germplasms.Among them,the germplasm resources between different catego-ries and subcategories with distant genetic relationships could be used as parents for hybrid breeding.
