摘要
目的研究人体骨骼肌失神经后肌细胞生成素的表达变化,了解失神经骨骼肌的再生状态。方法以11例正常与失神经不同时间人体骨骼肌为标本,提取总核糖核酸(RNA),通过半定量RT-PCR技术,检测失神经骨骼肌中肌细胞生成素(myogenin)mRNA的表达变化。结果人体骨骼肌失神经后,肌细胞生成素在1年内呈高水平表达,7个月达到顶峰,为正常骨骼肌表达的38倍,12个月时为21倍,至26个月下降至11倍。结论周围神经损伤后,应在伤后1年内尽早修复。
Abstract
Objective To detect the change of expression of myogenin in humanskeletal muscle after denervation and understand the regeneration of denervated skeletal muscle. Methods 11 cases of normal and denervated human skeletal muscle at different intervals were involved in the experiment. The total RNA was harvested . The change of expression of myogenin mRNA in human skeletal muscle after denervation was detected by semiquantative RT - PCR methods. Results The expression of myogenin persisted at high level within 1 year, with its peak at the seventh month which was 38 times as much as that of expression in normal skeletal muscle. It was 21 times at 12 month and decreased to 11 times at 26 month. Conclusions Repair should be done as early as possible within one year after injury of peripheral nerve.
