Effect of electroacupuncture antiferroptosis on lumbar spinal cord neurons in rats with peripheral nerve injury
Objective To study the regulatory effect of electroacupuncture on ferroptosis of corresponding spinal cord segment neurons in rats with peripheral nerve injury.Methods Sixty SPF grade rats were randomly divided into sham surgery group,model group,and electroacupuncture group,with 20 rats in each group.Except for the sham operated group rats,the other two groups of rats were established models of sciatic nerve injury.One day after modeling,electroacupuncture intervention was performed at the"Huantiao"and"Weizhong"acupoints,and samples were taken after 28 days of intervention.The functional changes of the sciatic nerve and the recovery rate of wet weight of the gastrocnemius muscle in ratswere observed,and the morphological and quantitative changes of neurons in the sciatic nerve and L4 to L6 spinal cord were detected using HE staining.PCR and Western Blot were used to observe the mRNA and protein expression of GPX4 in spinal cord tissue,and iron ion concentration changes in spinal cord tissue were detected using an iron ion concentration kit.Results Compared with the sham surgery group,all rats showed a decrease in sciatic nerve function and a decrease in wet weight recovery rate of the gastrocnemius muscle after modeling.After electroacupuncture intervention,compared with the model group,the sciatic nerve function index and wet weight recovery rate of the gastrocnemius muscle in the electroacupuncture group were significantly increased.Compared with the sham surgery group,the model group showed a decrease in GPX4 mRNA and protein expression in the spinal cord tissue,and an increase in iron ion concentration.Compared with the model group,the expression of GPX4 mRNA and protein in the spinal cord tissue of rats in the electroacupuncture group increased,while the concentration of iron ions decreased.Conclusion Electroacupuncture intervention can promote the recovery of neural function in rats with sciatic nerve injury,and at the same time,electroacupuncture has a protective effect on spinal cord neurons in the corresponding segment.The mechanism of action is related to the antiferroptosis on lumbar spinal cord neurons.
