Mechanism Analysis of Astragalus Rhubarb on Reducing Inflammation and Oxidative Stress in Mice with Sepsis Through SIRT3
Objective To explore and analyze the mechanism of Astragalus rhubarb on reducing inflammation and ox-idative stress in mice with sepsis through SIRT3.Methods A total of 50 8-week-old ICR grade female mice were selected to establish sepsis model by cecal ligation and perforation(CLP)method,and the mice were divided into sham operation group(NC group),model group(CLP group),Astragalus membranaceus group(HQ group),rhubarb group(DH group),and Astragalus membranaceus+Rhubarb group(HQ+DH group)by random table method.The serum levels of C-reactive pro-tein(CRP),procalcitonin(PCT),interferon-γ(IFN-γ),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),super-oxide dismutase(SOD),malondialdehyde(MDA),nitric oxide(NO)and SIRT3 were detected by enzyme-linked immu-nosorbent assay.Immunohistochemistry was used to detect SIRT3 protein expression,RT-PCR was used to determine SIRT3 mRNA level,and Western Blotting assay was used to determine SIRT3 protein expression level.Results Compared with NC group,serum levels of CRP,PCT,TNF-α,IFN-γ,IL-6,MDA and NO in CLP group were significantly increased(P<0.05).Compared with CLP group,HQ group,DH group and HQ+DH group had significant inhibitory effects on serum CRP,PCT,TNF-α,IFN-γ,IL-6,MDA and NO(P<0.05),among which HQ+DH group had the most significant inhibi-tory effect.Compared with NC group,the levels of serum SOD and SIRT3 in CLP group were significantly decreased(P<0.05).Compared with CLP group,HQ group,DH group and HQ+DH group had significant up-regulation effects on serum SOD and SIRT3(P<0.05),and HQ+DH group had the most significant up-regulation effect.HQ+DH group significantly up-regulated SIRT3 protein and mRNA expression levels(P<0.05).Conclusion Astragalus rhubarb can up-regulate SIRT3 protein expression in sepsis mice,significantly inhibit inflammatory response and index levels,and alleviate oxidative stress damage,which can provide a new idea for further research and development of innovative traditional Chinese medicine and technology.
