Effect and mechanism of Five-Flavor Sophora Flavescens Enteric-Coated Capsules on rats with ulcerative colitis
Objective To study the effect and mechanism of Five-Flavor Sophora Flavescens Enteric-Coated Capsules on rats with ulcerative colitis(UC).Methods The UC rat model was established by 3%dextran sulfate sodium(DSS).After successful modeling,the rats were randomly divided into blank group,model group,mesalazine group and Five-Flavor Sophora Flavescens Enteric-Coated Capsules group,with 10 in each group.The rats were given normal saline or corresponding drugs by intragastric administration every day for two consecutive weeks.Disease activity index(DAI)score and colonic mucosal damage index(CMDI)score were observed and compared among groups,and the expression levels of interleukin(IL-6)and interleukin(IL-10)in serum and expressions of IL-6 and IL-10 mRNA in colon tissues were detected by enzyme-linked immunosorbent assay(ELISA)and fluorescence quantitative reverse transcription polymerase chain reaction(qRT-PCR),respectively.The expression level of mucin 2(MUC2)and the activity of myeloperoxidase(MPO)in colon tissues were determined by myeloperoxidase colorimetry method,respectively.Results Compared with blank group,DAI score,CMDI score,serum IL-6 content,IL-6 mRNA expression in colon tissue,and MPO activity in model group were all significantly increased(all P<0.05),while IL-10 content,IL-10 mRNA expression in colon tissue,and average optical density(AOD)of MUC2 decreased significantly(all P<0.05).Compared with model group,DAI score,CMDI score,serum IL-6 content,IL-6 mRNA expression in colon tissue,and MPO activity of rats in mesalazine group and Five-Flavor Sophora Flavescens Enteric-Coated Capsules group were significantly decreased(all P<0.05),while the content of IL-10,IL-10 mRNA expression in colon tissue,and AOD value of MUC2 were significantly increased(all P<0.05).Conclusion Five-Flavor Sophora Flavescens Enteric-Coated Capsules may promote intestinal mucosal repair and alleviate inflammation in UC rats by decreasing the level of pro-inflammatory factor IL-6 and increasing the level of anti-inflammatory factor IL-10,up-regulating the expression of MUC2 and decreasing MPO activity.
