Effects of echinoside on Wnt/β-catenin signaling pathway in atherosclerotic vascular calcification mice
Objective To investigate the effect of echinoside(ECH)on the Wnt/β-catenin signaling pathway in mice with atherosclerotic(AS)vascular calcification.Methods Forty-eight 7-week-old male ApoE-/-mice were randomly divided into 6 groups,each with 8 mice:model group,ECH 12.5,25 and 50 mg/kg groups,Sta group,and control group,and they were given different treatment regimens.HE staining was used to observe the tissue ecological changes in the aorta of mice,and to detect the levels of biochemical indexes in the serum of mice.The expression level of aortic[3-catenin mRNA was detected by RT-qPCR,the expression levels of aortic β-catenin,Wnt3a,LRP5 and BMP-2 were detected by Western blotting,and the expression level of aortic LRP5 was detected by immunohistochemistry.Results The mem-branes in the ECH 25,ECH 50 mg/kg and Sta groups were significantly thinner than those of the model group,with aligned elastic fibres and significantly reduced plaques;while the levels of serum TG,TC,and LDL-C in the three groups were statistically lower compared with the control group(all P<0.01).HDL-C level in the ECH 12.5,25 and 50 mg/kg groups and Sta group was significantly higher than that of the model group(all P<0.001).In the model group,aortic β-catenin mRNA expression level was higher than that of the control group,while β-catenin mRNA expression level in the ECH 12.5,25,and 50 mg/kg groups and the Sta group was lower than that of the model group,differences of which were all statistically significant(all P<0.05).The protein expression levels of aortic β-catenin,Wnt3a,LRP5 and BMP-2 in the model group were higher than those of the control group,as well as ECH 25 and 50 mg/kg groups and Sta group,and the differences were statistically significant(all P<0.01).In the model group,aortic LRP5 protein expression level was statistically higher than that of the control group(P<0.05),as well as ECH 12.5,25,and 50 mg/kg groups and the Sta group(all P<0.01).Conclusion ECH effectively ameliorates the onset and development of AS calcification of ApoE-/-mice on a high-fat diet,the mechanism of which may be related to the regulation of Wnt/β-catenin signalling.
