Expression and clinical significance of IKBKE,YAP1 and TEAD2 in colorectal adenocarcinoma
Objective To explore the expression and clinical significance of inhibitor of nuclear factor kappa-B kinase subunit epsilon(IKBKE),Yes-associated protein 1(YAP1),and transcriptional enhancer activation domain 2(TEAD2)in colorectal cancer tissues(CRC).Methods Paraffin-fixed samples of CRC and adjacent tissues were obtained from 142 patients at the First Affiliated Hospital of Bengbu Medical University from January 2016 to December 2017.Protein expressions of IKBKE,YAP1,and TEAD2 were detected by immunohistochemistry.The correlation of the expression of the three proteins in CRC tissues,and the relationship of positive expression rate of the proteins and clinicopathological parameters with prognosis were analyzed.The Kaplan-Meier survival curves were plotted to compare the survival differences of patients with different expression conditions of these proteins.Cox regression was used to analyze the risk factors affecting patient outcomes.Results Positive expressions of IKBKE,YAP1 and TEAD2 in CRC tissues were significantly higher than those in adjacent normal tissues(65.5%vs.9.9%,73.9%vs.14.1%,and 66.9%vs.8.5%,respectively;all P<0.05).The expressions of IKBKE,YAP1,and TEAD2 were all positively correlated with the degree of tumor differentiation,depth of invasion,lymph node metastasis,and tumor-node-metastasis(TNM)stage;while YAP1 and TEAD2 expressions were also positively correlated with distant metastasis.Spearman analysis showed that IKBKE expression was positively correlated with YAP1 and TEAD2 expressions in CRC tissues(both P<0.01).Kaplan-Meier survival analysis showed reduced overall survival for patients with positive expressions of the three proteins.Cox regression analysis revealed that positive expressions of IKBKE,YAP1,and TEAD2,higher degree of tumor differentiation,and higher TNM stage were also independent risk factors for the prognosis of CRC patients.Conclusion The positive expressions of IKBKE,YAP1 and TEAD2 in CRC are correlated with factors such as tumor differentiation,TNM stage,and metastasis.Testing the expression levels of the three proteins may provide potential targets for colorectal cancer treatment and help to evaluate the prognosis.
