Analysis on the characteristics of serum androgen levels in women of reproductive age in Hangzhou based on mass spec-trometry
Objective To detect and analyze the characteristics of six serum androgens in 20-40 year-old women in Hangzhou,Zhejiang province by liquid chromatography-tandem mass spectrometry(LC-MS/MS).Methods Healthy women of reproductive age from November 2021 to February 2022 were recruited from Zhejiang Provincial People's Hospital,Affiliated Hangzhou First People's Hospital of School of Medicine,Westlake University,and Tongde Hospital of Zhejiang Province following inclusion and exclusion criteria.A total of 380 women aged 20-40 years were selected as subjects.Their serum levels of total testosterone(TT),free testosterone(FT),dihydrotestosterone(DHT),androstenedione(A4),dehydroepiandrosterone(DHEA)and dehydroepiandrosterone sulphate(DHEAS)were determined by LC-MS/MS.The percentile method(P25-P97.5)was used to establish the distribution intervals and the results were stratified for analysis.In addition,the relationship between six androgens and metabolic parameters was analyzed.Results The distribution intervals for TT,FT,DHT,A4,DHEA and DHEAS of the total population(20-40 years old)were 81.77-477.21,1.00-4.05,50.00-157.28,550.33-2 172.06,1 675.51-12 054.90 pg/mL and 0.92-4.05 μg/mL,respectively.TT,FT and DHT distribution intervals(P97.5)were significantly lower than those from the Mayo laboratory.When stratified by age,the differences in six serum androgens between the 20-30-and 31-40-year-old groups were statistically significant(P<0.05),but the density curve overlaps highly.When stratified into the early-middle follicles phase,late follicular phase and luteal phase,no significant differences in DHT,DHEA,or DHEAS were observed(all P>0.05),while TT,FT and A4 showed phase differences(all P<0.05),but the density curve overlaps highly.In addition,six serum androgens and sex hormone-binding globulin were correlated with glucose and lipid metabolism parameters in healthy women of reproductive age with normal BMI(all P<0.05).Conclusion The serum androgen levels in women of reproductive age in Hangzhou is significantly different from those in North American population.Therefore,local reference ranges should be established for clinical application.
