SOX14对膀胱癌细胞生物学行为的影响

目的 观察性别决定区Y框基因转录因子(SOX)14在膀胱癌组织中的表达情况,探讨其对膀胱癌细胞增殖、迁移和侵袭等生物学行为的影响.方法 收集2019年12月至2022年6月温州医科大学附属第一医院手术切除的膀胱癌组织(37例)和癌旁组织(14例),采用qRT-PCR法检测癌组织和癌旁组织中、膀胱癌细胞和正常尿路上皮细胞中SOX14 mRNA的表达水平.使用过表达和敲低SOX14载体转染膀胱癌细胞,采用细胞计数试剂盒(CCK)-8法、细胞划痕实验和Transwell实验检测并比较不同转染细胞增殖、迁移和侵袭能力的差异.结果 膀胱癌组织中SOX14 mRNA相对表达量显著低于癌旁组织,5637、T24、EJ、J82和RT4等5种膀胱癌细胞中SOX14 mRNA相对表达量均显著低于正常尿路上皮细胞.相比相应对照组,过表达SOX14后膀胱癌细胞存活率、细胞迁移和侵袭能力均显著降低;敲低SOX 14后膀胱癌细胞存活率、细胞迁移和侵袭能力均显著升高.结论 SOX14在人膀胱癌组织和细胞中低表达.调控SOX14mRNA的表达水平能影响膀胱癌细胞的增殖、迁移和侵袭能力.提示SOX14可能是治疗膀胱癌的新靶点.
Effect of SOX14 on biological behaviour in bladder cancer cells
Objective To investigate the expression of sex determination region of Y chromosome-box transcription factor 14(SOX14)in bladder cancer and its effects on the cell proliferation,migration and invasion on the biological behavior of bladder cancer.Methods Thirty seven bladder cancer specimens and 14 adjacent bladder tissue specimens were collected in the First Affiliated Hospital of Wenzhou Medical University from December 2019 to June 2022.The expression of SOX14 in tissue specimens were detected by qRT-PCR.The expression of SOX14 in bladder cancer cells,and normal urothelial cells were also detected.The SOX14 gene was transfected or knocked down in bladder cancer cells to construct SOX14 overexpression or knock down cell lines.The effects of SOX14 on the cell proliferation,migration and invasion were investigated by CCK-8 text,wound healing and Transwell assay.Results The relative expression of SOX14 mRNA in bladder cancer tissues was significantly lower than that in adjacent tissues.The relative expression of SOX 14 mRNA in bladder cancer 5637,T24,EJ,J82,RT4 cells was significantly lower than that in normal urothelial cells.After overexpression of SOX14 in bladder cancer cells,the cell survival rate was lower than that of control cells,and the migration and invasion ability were significantly decreased.After knocking down SOX14,the survival rate of bladder cancer cells increased,and the migration and invasion ability of cells were significantly enhanced compared with the control group.Conclusion The relative expression of SOX14 is low in human bladder cancer.Overexpression of SOX14 mRNA inhibits the proliferation,migration and invasion of bladder cancer cells;while knocking down SOX14 mRNA promotes cell proliferation,migration and invasion.
