Effect of SOX14 on biological behaviour in bladder cancer cells
Objective To investigate the expression of sex determination region of Y chromosome-box transcription factor 14(SOX14)in bladder cancer and its effects on the cell proliferation,migration and invasion on the biological behavior of bladder cancer.Methods Thirty seven bladder cancer specimens and 14 adjacent bladder tissue specimens were collected in the First Affiliated Hospital of Wenzhou Medical University from December 2019 to June 2022.The expression of SOX14 in tissue specimens were detected by qRT-PCR.The expression of SOX14 in bladder cancer cells,and normal urothelial cells were also detected.The SOX14 gene was transfected or knocked down in bladder cancer cells to construct SOX14 overexpression or knock down cell lines.The effects of SOX14 on the cell proliferation,migration and invasion were investigated by CCK-8 text,wound healing and Transwell assay.Results The relative expression of SOX14 mRNA in bladder cancer tissues was significantly lower than that in adjacent tissues.The relative expression of SOX 14 mRNA in bladder cancer 5637,T24,EJ,J82,RT4 cells was significantly lower than that in normal urothelial cells.After overexpression of SOX14 in bladder cancer cells,the cell survival rate was lower than that of control cells,and the migration and invasion ability were significantly decreased.After knocking down SOX14,the survival rate of bladder cancer cells increased,and the migration and invasion ability of cells were significantly enhanced compared with the control group.Conclusion The relative expression of SOX14 is low in human bladder cancer.Overexpression of SOX14 mRNA inhibits the proliferation,migration and invasion of bladder cancer cells;while knocking down SOX14 mRNA promotes cell proliferation,migration and invasion.
