Construction of a prognostic model for colon cancer based on bioinformatics analysis of ferroptosis-related genes
Objective To screen colon cancer(CC)prognosis and ferroptosis related genes(CFRGs)and construct CC prognostic model.Methods Relevant data were obtained from The Cancer Genome Atlas(TCGA)and FerrDb database,and CFRGs were screened by bioinformatics method.The expression difference of the selected CFRGs was analyzed by TCGA database.Kaplan-Meier curve and time-ROC curve were used to analyze the prognosis of the selected CFRGs.The CFRGs Nomogram model was constructed by univariate and multivariate Cox regression analysis,and the predictive ability of the model was verified by Calibration analysis.Results Three differentially expressed ferroptosis-related genes ALOXE3,CRYAB and CDKN2A were identified;the expression of ALOXE3 and CDKN2A was upregulated and CRYAB expression was downregulated.CC patients with higher expression of ALOXE3,CRYAB and CDKN2A had a poorer survival outcome.A risk scoring system of three genes was constructed,risk score=0.172 × ALOXE3(exp)+0.126 × CRYAB(exp)+0.171 × CDKN2A(exp)-0.772,and the AUC for predicting 1,3 and 5-year survival of CC patients was 0.642,0.643 and 0.593,respectively.The CFRGs Nomogram model was constructed with age,sex and risk score,and the prediction ability was good(C index was 0.651).Calibration analysis showed that the model predicted 1-,3-and 5-year survival rates were in good agreement with the ideal line segment.Conclusion The survival prognosis of colon cancer patients is significantly associated with ALOXE3,CRYAB and CDKN2A,and the constructed risk scoring system of three genes and CFRGs Nomogram model combined with age,sex and risk score have good efficacy.
