Correlation between UCH-L1 and PD-L1 expression in non-small cell lung cancer and their clinicopathological significance
Objective To investigate the correlation between ubiquitin carboxy terminal hydrolase 1(UCH-L1)and programmed cell death ligand 1(PD-L1)expression and their clinicopathological significance in non-small cell lung cancer(NSCLC).Methods A total of 105 paraffin embedded NSCLC samples and 12 paired NSCLC tissue and adjacent normal tissue samples were retrospectively collected from Affiliated Hangzhou First People's Hospital,School of Medicine,Westlake University from January 2018 to December 2019.The expression of UCH-L1 and PD-L1 in 12 fresh paired NSCLC and adjacent normal tissue samples and in 105 paraffin embedded NSCLC samples was detected by Western blot and En Vision two-step immunohistochemistry,respectively.The association between UCH-L1 and PD-L1 protein expression,clinico-pathological features,and prognosis of NSCLC tissues,as well as the correlation between the two protein expression were analyzed.Results The results showed that among the 12 patients,the level of UCH-L1 protein expression in fresh NSCLC tissues was significantly higher than that in adjacent normal tissues in 9 cases(except cases 6,9,and 12),while the level of PD-L1 protein expression was significantly lower than that in adjacent normal tissues in 10 cases(except cases 2 and 11).The positive rate of UCH-L1 and PD-L1 in 105 cases of NSCLC was 28.6%(30/105)and 38.1%(40/105),respectively.UCH-L1 expression was associated with tumor type,tumor size and TNM stage(all P<0.05),while PD-L1 expression was just associated with tumor type(P=0.001).The NSCLC patients with positive UCH-L1 expression had a worse progression-free survival(PFS)rate than those with negative expression(P<0.001),while there was no significant difference in PFS rate between patients with positive and negative PD-L1 expression(P=0.055).Pearson correlation analysis revealed that UCH-L1 expression was positively associated with PD-L1 expression(r=0.329,P=0.001).Conclusion UCH-L1 acts as a tumor gene in the occurrence and development of NSCLC.The expression of UCH-L1 and PD-L1 is positively correlated,and the two may have a synergistic effect and coordinately participate in the occurrence and development of NSCLC.
