Expressions of ribosomal protein L34 and STAT3 in cervical cancer tissue and their relationships with clinicopathologic features and prognosis
Objective To investigate the expressions of ribosomal protein L34(RPL34)and signal transduction and transcription regulator 3(STAT3)in cervical cancer tissue and their relationships with clinicopathologic features and prognosis.Methods Cervical cancer tissues and normal cervical tissues(as control)were collected from Taizhou Central Hospital from January 2019 to June 2022,each from 30 patients;Paraffin specimens of 76 cases of cervical cancer and 40 normal cervical tissues(as control)were also collected from January 2017 to June 2022.The relative expressions of RPL34 and STAT3 mRNA in fresh tissue were detected by qRT-PCR,while the expressions of RPL34 and STAT3 in paraffin specimens were detected by immunohistochemistry so as to analyze the expression differences of RPL34 and STAT3 among cervical cancer patients with different clinicopathological characteristics.The correlation between RPL34 and STAT3 expressions was analyzed using Spearman rank correlation analysis,and Kaplan-Meier survival curves were drawn to analyze the prognostic significance of RPL34 and STAT3 expressions.Results The RPL34 mRNA relative expression and protein expression in cervical cancer tissue were significantly lower than those in normal control tissue(both P<0.01),while the STAT3 mRNA relative expression and protein expression were significantly higher than those in normal control tissue(both P<0.01).There were statistically significant differences in the expression level of RPL34 in cervical cancer patients with different International Federation of Gynecology and Obstetrics(FIGO)stages,depth of invasion and lymph node metastasis(all P<0.01),and statistically significant differences in the expression level of STAT3 in cervical cancer patients with different FIGO stages,depth of invasion,lymph node metastasis and differentiation(all P<0.01).The expression of RPL34 in cervical cancer tissue was negatively related to the expression of STAT3(P<0.01).Compared with patients with negative expression,those with positive RPL34 expression had a significantly higher 3-year overall survival rate after surgery(P<0.01),while those with positive STAT3 expression showed a significantly lower 3-year overall survival rate after surgery(P<0.01).Conclusion RPL34 mRNA and protein in cervical cancer tissue are down-regulated,while STAT3 mRNA and protein are up-regulated.The expressions of RPL34 and STAT3 are related to the clinicopathological characteristics including the depth of infiltration and lymph node metastasis of cervical cancer and so on,and they affect the 3-year overall survival rate of patients.RPL34 and STAT3 are expected to become prognostic markers for cervical cancer.
