Antibacterial activity of prodigiosin against methicillin-resistant Staphylococcus aureus isolated from wounds and its mechanism
Objective To investigate the antibacterial effect and its mechanism of prodigiosin on methicillin-resistant Staphylococcus aureus(MRSA)isolated from wounds.Methods A total of 64 strains of MRSA isolated from wound tissue infections of the First Hospital of Jiaxing from 2021 to 2022 were selected.The minimum inhibitory concentrations(MIC)of prodigiosin used alone and combined with levofloxacin were both determined by the micro-broth dilution method,and the fractional inhibitory concentration was calculated to evaluate the bacteriostatic effect of the combined use.The antibacterial mechanism of prodigiosin was observed by transmission electron microscopy.Results The 50%MIC and 90%MIC of prodigiosin against MRSA were 0.5 and 1.0 mg/L,respectively.The synergistic effect of prodigiosin and levofloxacin was observed in one strain(1.54%),and the additive effect was in 30 strains,accounting for 46.15%.It was observed by transmis-sion electron microscopy that prodigiosin had a significant effect on destroying the integrity of the cell membrane structure of MRSA strains.Conclusion Prodigiosin can inhibit MRSA by destroying the bacterial cell membrane and has a certain synergistic or additive effect when combined with levofloxacin.
