Analysis of Bioactive Compounds,Antioxidant and Antibacterial Activity of Different Polar Parts of Rhubarb
[Objective]To investigate the content of active ingredients in different polar parts of rhubarb and antioxidant and antibacterial activities in vitro.[Methods]Ninety percent ethanol reflux was used to extract rhubarb,and chloroform,ethyl acetate and n-butanol were used to extract different polar parts;anthraquinone compounds were determined by high performance liquid chromatography(HPLC);total phenol content(TPC)and total flavonoid content(TFC)were detected by UV spectrophotometry;1,1-diphenyl-2-picrylhydrazyl(DPPH)method,2,2'-azinobis-(3-ethylbenzthiazoline-6-sulphonate)(ABTS)method,and total reducing power method were used to determine antioxidant activity.The effects of different polar parts of rhubarb on Escherichia coli,Staphylococcus aureus and Bacillus subtilis were measured by the diameter of inhibition zone and growth curve.[Results]The contents of aloe emodin-8-O-glucoside and rhein-8-O-glucoside were the highest in n-butanol part,with peak areas of(2.72±0.07)mAU*min and(2.59±0.06)mAU*min,respectively.TPC and TFC were the highest,with content of(29.95±4.95)mg·g-1 and(35.71±3.26)mg·g-1,respectively.The antioxidant and antibacterial activities of the n-butanol fraction were the strongest;antioxidant activity was positively correlated with aloe emodin-8-O-glucoside,rhein-8-O-glucoside,TPC and TFC(P<0.05).[Conclusion]The n-butanol fraction of rhubarb extract has the strongest antioxidant and antibacterial activity,which may be related to aloe emodin-8-O-glucoside and rhein-8-O-glucoside,and the results has guiding significance for further research.
