Study on the Intervention of Panax Ginseng Saponin Serum Exosomes on Inflammatory Injury in Rats with Intracerebral Hemorrhage
Objective:To study the effects of Panax ginseng saponin serum exosomes on pathologi-cal morphological changes in brain tissue and expression of inflammatory factors in rats with Intra-cerebral Hemorrhage(ICH).Explore the protective effect on brain tissue inflammatory injury in ICH rats.Methods:Thirty successfully modelled rats were divided into Panax ginseng saponin se-rum exosome group,Panax ginseng saponin group,and model group by random number table meth-od,and were given Panax ginseng saponin serum exosome(20μg/day),Panax ginseng saponin(100mg/kg),and equal volume of physiological saline for tail vein injection for 10 days consecu-tively,meanwhile a normal rat group was set up for comparison.After the intervention,compare the degree of histopathological damage in the brain tissue of the four groups of rats.RT-PCR was used to detect the mRNA expression levels of inflammatory indexes NF-κB,NLRP3,IL-6 and TNF-α in the tissues surrounding the intracerebral hemorrhage injury.The levels of NF-κB,IL-6 and IL1β in homogenates of brain tissues were detected by using ELISA.Results:Compared with the model group,the Panax ginseng saponin serum exosome group and Panax ginseng saponin group had dif-ferent degrees of intracerebral hemorrhage resorption and reduction of inflammatory infiltration,and the effect of Panax ginseng saponin serum exosome group was better than that of Panax ginseng sa-ponin group.The expression levels of inflammatory indexes NF-κB,NLRP-3,IL-6,TNF-α and IL1β in Panax ginseng saponin serum exosome group were significantly down-regulated compared with that in model group and lower than that in Panax ginseng saponin group,and the difference was statistically significant(P<0.05).Conclusion:Panax ginseng saponin serum exosome as a bioactive substance can effectively improve the level of brain tissue inflammation after intracerebral hemor-rhage in intracerebral hemorrhage rats,alleviate pathological damage,and have a certain protective effect on intracerebral hemorrhage injury.
